DNAおよびRNAのメチル化は、遺伝子制御、ゲノム安定性、RNA代謝、および疾患の進行に関与する重要なエピジェネティック(後成的)およびエピトランスクリプトミクス(転写後)修飾です。セルフリーDNAやRNAにおける異常なメチル化パターンは、がんの検出、疾患のモニタリング、および治療方針の決定(層別化)に有用なバイオマーカーとして注目されています。
しかし、バイサルファイトシーケンシング、メチル化特異的PCR、MeRIP-seq、SCARLET、LC-MS/MSといった従来の手法は、過酷な処理や多量のサンプルを必要としたり、装置が複雑であったり、あるいは部位特異的な分解能が不足していたりすることが多く、臨床現場やポイントオブケア(POC)での応用が制限されてきました。
近年のCRISPRを用いた診断技術は、プログラム可能な核酸認識と、標的核酸下で細胞内の核酸をトランス切断するコラテラル活性を介したシグナル増幅を組み合わせることで、有望な代替手段を提供します。本総説では、DNAおよびRNAメチル化を検出するための最新のCRISPR戦略について概説されています。
具体的には、化学的変換を併用するアッセイ、制限酵素を介した検出、メチル化によって制御されるCas活性を利用した増幅不要の直接センシング、酸化シトシン誘導体の検出、逆転写を介したCas12によるm6A検出、および構造感受性Cas13センシングなどが含まれています。
また、メチル化依存的な配列変換、酵素のアクセス性、ポリメラーゼの挙動、核酸構造といった要素が、いかにしてCRISPRで読み取り可能なシグナルへと変換され得るかについても詳述されています。
最後に、ポイントオブケアでのメチル化診断、DNA・RNA統合プロファイリング、改変型Casエフェクター、AIを活用したアッセイ設計、およびCRISPR適合型メチローム解析といった分野における、現在のトランスレーショナルな課題と新たな展望について議論されています。
